摘要:为了筛选适用于酿酒生产的产香酵母菌,以花果香高温大曲为实验材料,采用平板稀释涂布法和划线分离法得到6株菌株。通过感官评价和产酯透明圈的初筛实验,结合固态发酵产酯能力的复筛实验,得到1株产香能力较强的菌株Y2。结合形态学分析和分子鉴定结果表明,酵母菌Y2为扣囊复膜酵母(Saccharomycopsis fibuligera)。耐受性测定显示酵母菌Y2可较好地耐受乙醇浓度8%、pH 3.0的酸性环境和43℃的环境。

Study on the Screening,Identification and Property of Aroma-producing Yeasts in High-temperature Daqu

Abstract: In order to screen aroma-producing yeast suitable for brewing production, high-temperature daqu with flower and fruit aroma was used as experimental material, 6 strains were isolated by dilution plate method and streak separation method. Through primary screening experiment of sensory evaluation and ester-producing transparent circle, combined with the rescreening experiment of solid fermentation ester-producing ability, the strain Y2 with good aroma- producing ability was obtained. Combined with the morphological analysis and molecular identification result, showing Y2 yeast was Saccharomycopsis fibuligera. The tolerance determination showed that Y2 yeast could tolerate the environment of 8% ethanol concentration, acidic environment of pH 3.0 and 43℃ better.
