摘要:为提升烟叶提取液香气特征,该研究从清香型大曲中分离筛选高产乙酸乙酯的优良酵母菌,并考察其在烟叶提取液发酵中的应用效果。结果表明,从清香型大曲中初筛获得9株菌株,以乙酸乙酯和生长性能为指标,复筛确定SMR-Y-6为高产乙酸乙酯酵母菌,并鉴定为酿酒酵母(Saccharomyces cerevisiae)。采用单因素和正交实验优化烟叶提取液液态静置发酵条件,最优条件为:接种量4%、发酵温度29℃、发酵时间4d、初始pH值7.0,发酵后乙酸乙酯含量为1 739.88 mg/L,相比发酵前增加130.55%。此外,经GC-MS结果证实,该菌株可提升85.8%挥发性致香成分相对含量(乙酸乙酯除外),增加19.1%挥发性致香成分种类。该研究结果表明SMR-Y-6酿酒酵母可作为烟叶发酵潜在优良菌种应用于工业生产。

Screening of Saccharomyces cerevisiae with High Yield of Ethyl Acetate from Light Flavor Daqu and Study on Its Fermentation Effect on Tobacco Extract

Abstract: In order to improve the aroma characteristics of tobacco extract, this study isolated and screened the good yeast with high yield of ethyl acetate from light flavor daqu, and investigated its application effect in fermentation of tobacco extract. The results showed that 9 strains were obtained by preliminary screening from light flavor daqu, by using ethyl acetate and growth performance as indicators, SMR-Y-6 was determined as yeast with high yield of ethyl acetate by rescreening, and identified as Saccharomyces cerevisiae. Single factor and orthogonal experiments were used to optimize the liquid static fermentation conditions of the tobacco extract. The optimal conditions were as follows: 4% inoculation amount,29 ℃ fermentation temperature,4 d fermentation time, initial pH value 7.0, after fermentation the ethyl acetate content was 1 739.88 mg/L, which increased by 130.55% compared to before fermentation. In addition, GC-MS results confirmed that the strain could increase the relative content of volatile aroma components (except ethyl acetate) by 85.8%, and increase the types of volatile aroma component by 19.1%. The research results showed SMR-Y-6 Saccharomyces cerevisiae could be used in industrial production as a potential good strain for tobacco fermentation.
